The ImagEM X2, a completely redesigned camera featuring a back-thinned EM-CCD sensor, offers maximum speed and precision performance for low-light imaging.

The ImagEM X2 makes superfast exposures possible and has the sensitivity to provide visually pleasing and quantitatively meaningful images in a photon-starved environment. It delivers 70 frames/s at the full resolution of 512 x 512 pixels with a high signal-to-noise ratio, enabling quantitative high-speed, low-light imaging. When binning or a region of interest is selected, this new camera produces images at even higher frame rates (up to 1076 frames/s).

Additional features allow for optimized camera triggering and streamlined connectivity through IEEE 1394b. The ImagEM X2 also has improved overall signal-to-noise ratio, increased non-EM dynamic range, and built-in EM gain measurement and calibration functions. It also features a software-controllable shutter that prevents EM gain degradation and image lag. To prevent EM gain degradation, the shutter is closed when the built-in EM gain protection feature is enabled. In addition, closing the shutter when a user replaces a lens, for example, can prevent image lag.

The Fragment Analyzer™ by Advanced Analytical is a fluorescence-based capillary electrophoresis instrument for both sizing and quantifying nucleic acids (DNA and RNA).

Advanced Analytical Technologies

By using a sensitive intercalating dye coupled with a powerful LED light source, Fragment Analyzer™ Automated CE System obviates the need for fluorescent labeled primers and can be used to separate dsDNA fragments and RNA (mRNA and total RNA). The Fragment Analyzer™ Automated CE System is the most flexible system on the market today, with the greatest sensitivity, the highest separation resolution over a wide range, the widest dynamic range and the fastest separation times.

This instrument comes available for customers who wish to run either 12 samples or 96 samples at a time.

Currently kits are available to:

• quantify and qualify NGS fragments.
• quantify and qualify RNA.
• quantify and qualify genomic DNA.
• do Mutation Detection, also known as TILLiNG, a reverse genetics technique.

The application kits help laboratories to streamline workflow and avoid bottlenecks typically found with competitive lab-on-a-chip methods. Gels formulated to provide high resolution separation are also available for general fragment analysis, such as SSR/microsatellites, small and large PCR fragments (amplicons) and resolve fragments sized between 10 bp and 40,000 bp with resolution down to 2bp.

Each and every kit is performance-tested prior to shipment to ensure quality, accuracy and ease of use for customers. Most kits require a single pipetting step before placement of the sample plate onto the instrument.

An analytical software package, PROSize™, greatly reduces post-separation analysis time. Put them all together — the instrument itself, the QC-tested application kits, and the analytical software — and the combination makes the Fragment Analyzer™ Automated CE System a powerful package for reducing labor and improving lab workflow and throughput.

His Mag- and Ni Sepharose excel are immobilized metal ion affinity chromatography (IMAC) media (resins) designed for capture and purification of histidine-tagged proteins secreted into eukaryotic cell culture supernatants.

Nickel ions are very strongly bound to both media, enabling direct loading of large sample volumes without removing agents that normally would cause metal ion stripping. Both media significantly simplify and speed up the workflow and are excellent choices when working with purification of histidine-tagged proteins secreted from eukaryotic cells.

Background:

IMAC purification of histidine-tagged proteins secreted into eukaryotic cell culture supernatants has traditionally been challenging because of incompatibility between the IMAC medium and the cell culture medium. The cell culture medium strips the immobilized metal ions from the IMAC medium during sample loading, resulting in low or no binding of the target protein. The purification has been further complicated by the fact that the target protein concentration is often low. This requires the use of large sample volumes, which in turn might lead to increased metal ion stripping. Considerable sample pretreatment has been required to overcome these problems, an example being buffer exchange by diafiltration in combination with concentration procedures. Such pretreatment is time consuming, potentially harmful to sensitive proteins, and can cause unnecessary loss of target protein.

The new product can be used to quickly and reliably screen hundreds of plates from limited dilutions via automated image capture and analysis.
