His Mag Sepharose™ Ni

His Mag Sepharose Ni products are magnetic beads designed for efficient, small-scale purification/screening of histidine-tagged proteins from different sources. Histidine-tagged proteins are captured using immobilized nickel ions followed by collection of the beads using a magnetic device. You can use magnetic beads to simplify sample handling during small-scale purification.

His Mag Sepharose Ni delivers:

The beads are available in three pack sizes: 2 × 1 ml 5% medium slurry, 5 × 1 ml 5% medium slurry, and 10 × 1 ml 5% medium slurry—a 1 ml 5% medium slurry is the same as 50 μl sedimented medium. A 1 ml medium slurry is sufficient for 5 purification runs according to the recommended protocol.

Together with MagRack 6, a separation tool for handling the beads in microcentrifuge tubes, up to six samples can be processed in parallel. You can easily screen a larger number of samples in parallel with high throughput on a robotic device.


Simplified handling:

The magnetic bead format has excellent properties for small-scale experiments. The high density of the beads allows rapid capture by magnetic devices while the visibility of the beads ensures reliable collection of the bound histidine-tagged proteins in the purification procedure. The characteristics of the media are summarized in Table 1. All the products are provided with protocols optimized for purification of histidine-tagged proteins.

The µDAWN™  Multi-Angle static Light Scattering (MALS) detector performs absolute characterization of macromolecules eluting from UHPLC, determining molar mass and size independently of retention time and molecular reference standards. This instrument offers superb sensitivity over a wide range of molar mass, size and concentrations. In most respects the µDAWN is similar to the well-established miniDAWN™ TREOS™, a workhorse MALS instrument used in hundreds of labs around the world for characterizing proteins and other molecules smaller than 50 nm in radius.

Distinctly from the miniDAWN, the µDAWN is uniquely suitable for use in conjunction with UHPLC Size Exclusion Chromatography (µSEC-MALS) thanks to its small interdetector dispersion. In some cases it may be used with ion-exchange or other types of chromatography. Regardless of the specifics, the µDAWN produces minimal band broadening in order to maintain the narrow peaks typical of UHPLC.

The µDAWN and its companion Optilab® UT-rEX™ Refractive Index Detector interface to most standard UHPLC systems. As these instruments are based on the advanced technology of the miniDAWN TREOS and Optilab T-rEX, respectively, users may be confident in the underlying capabilities cited in thousands of peer-reviewed publications

The µDAWN may also be used in microbatch (off-line) mode or in conjunction with a Calypso II CG-MALS system for characterization of macromolecular interactions: affinity and absolute stoichiometry, label-free and immobilization-free, as well as second virial coefficients, cross virial coefficients, average molar masses and sizes.

Today, embryonic stem cells (ESC) and induced pluripotent stem cells (iPS) represent the most significant types of pluripotent stem cells (PSC).

In the beginning, ESC were cultured on feeder cell layers, primarily murine embryonic fibroblasts (MEF), in non-defined culture media containing fetal calf serum (FCS). Later on, FCS would be replaced by somewhat more defined serum replacements and the introduction of extracellular matrix (ECM) coatings supporting self-renewal of ESC allowed for feeder-free culture using feeder-cell conditioned medium. However, the culture process remained time-consuming, laborious and poorly defined.

In recent years PSC-culture has experienced significant technical progress. Chemically defined media compositions became a standard, followed by the first xeno-free defined formulations. However, all these culture systems still depended on non-defined, xenogenic and poorly standardized ECM preparations – a contradiction in terms when using defined / xeno-free media. Importantly, research recently overcame this hurdle, identifying natural as well as synthetic ECM molecules and peptides that support undifferentiated PSC-expansion. Scientists are now capable of completely defined / humanized derivation and culture of hPSC.

Despite these recent technical advancements, the established hPSC culture systems still share unfavourable properties. Most of them use supra-physiologically high amounts of growth factors and / or contain substances purified from human or animal origin. In addition animal-derived and / or non-defined ECM is used. The Cadence Inline Concentrator simplifies concentrating solutions of biomolecules to an in-line, single-use, ready-to-use process.

The recirculation loop associated with tangential flow filtration (TFF) conventionally used for this process is eliminated. Pump and tube sizes are reduced, product recovery is maximized, and sensitive proteins are exposed to less shear. The Cadence Single-Use Inline Concentrator moves the product being processed through a proprietary design flow path in a pre-assembled module with all connections already in place and designed to couple to available standard tubing manifolds.

Typical applications include monoclonal antibodies, fractionated antibodies, recombinant proteins, other proteins and recombinant blood factors. The Cadence Single-Use Inline Concentrator is used to concentrate clarified bulk after fermentation, and to reduce the number or size of bags needed and increase product concentration to improve chromatography performance.

Beckman Coulter Life Sciences is expanding their flow cytometry portfolio with the addition of 14 new human monoclonal antibodies from Trillium Diagnostics LLC.  The analyte specific reagents (ASRs) provide high specificity, convenience and flexibility in flow cytometry.  A new cell permeabilization reagent is also being offered. 

The new monoclonals include human anti-hemoglobin F, which recognizes a fetal hemogloblobin molecule; CD163 markers for anti-inflammatory signal and disease process research and CD64 markers, which react with Fc gamma receptors. Characterized, quality controlled, convenient, liquid reagents allow users to spend less time in sample preparation and more time on experiments.

Rounding out the line is a monoclonal for MNDA (myeloid nuclear differentiation antigen). All antibodies are available unconjugated or conjugated to a selection of fluorochromes. Intra-Cell™ concentrate for cell permeabilization is also being released.

Beckman Coulter Life Sciences offers an extensive line of human and non-human antibodies as well as a range of supporting reagents and immune monitoring products. The new antibodies are supplied in 1.0 mL of buffer, are ready to use and facilitate flexibility in experimental design.

“Convenience and specificity are important features for scientists,” said Mario Koksch, vice president and general manager for Beckman Coulter Life Sciences’ Flow Cytometry Business Unit. “These new antibodies are directed at important targets and will improve accuracy for researchers.”

By using a sensitive intercalating dye coupled with a powerful LED light source, Fragment Analyzer™ Automated CE System obviates the need for fluorescent labeled primers and can be used to separate dsDNA fragments and RNA (mRNA and total RNA). The Fragment Analyzer™ Automated CE System is the most flexible system on the market today, with the greatest sensitivity, the highest separation resolution over a wide range, the widest dynamic range and the fastest separation times.

This instrument comes available for customers who wish to run either 12 samples or 96 samples at a time.

Gels formulated to provide high resolution separation are also available for general fragment analysis, such as SSR/microsatellites, small and large PCR fragments (amplicons) and resolve fragments sized between 10 bp and 40,000 bp with resolution down to 2bp.

Each and every kit is performance-tested prior to shipment to ensure quality, accuracy and ease of use for customers. Most kits require a single pipetting step before placement of the sample plate onto the instrument.

An analytical software package, PROSize™, greatly reduces post-separation analysis time.
