Also by contrast with DNA transfection, mRNA delivery does not enter the nucleus, rather only cytosolic delivery is required, resulting in more a rapid expression method.

The Cadence Inline Concentrator simplifies concentrating solutions of biomolecules to an in-line, single-use, ready-to-use process.

The recirculation loop associated with tangential flow filtration (TFF) conventionally used for this process is eliminated. Pump and tube sizes are reduced, product recovery is maximized, and sensitive proteins are exposed to less shear. The Cadence Single-Use Inline Concentrator moves the product being processed through a proprietary design flow path in a pre-assembled module with all connections already in place and designed to couple to available standard tubing manifolds.

Typical applications include monoclonal antibodies, fractionated antibodies, recombinant proteins, other proteins and recombinant blood factors. The Cadence Single-Use Inline Concentrator is used to concentrate clarified bulk after fermentation, and to reduce the number or size of bags needed and increase product concentration to improve chromatography performance. 

Cost projections with BioSolve software show that, compared to conventional single-use TFF, the Cadence Single-Use Inline Concentrator provides a 31% cost of goods sold and a 66% capital investment savings per batch because of the elimination of the need for intermediate tanks, ability to use smaller chromatography columns, energy savings, and the ability to couple processes to reduce downtime.

High-efficiency CRISPR genome-editing tools for multiplex editing

The GeneArt® CRISPR Nuclease mRNA is wild type Cas9 as mRNA, for genome editing using CRISPR-Cas9 technology. Ready-to-transfect Cas9 mRNA circumvents the need for time-consuming cloning steps required when using CRISPR vector systems. Cas9 mRNA can be co-transfected with in vitro transcribed guide RNA (IVT gRNA) or a synthetic gRNA expression cassette containing U6 promoter. The gRNA expressing cassette and IVT gRNA template can be ordered as GeneArt® CRISPR Strings™ DNA, a 500 bp DNA fragment. Following transfection, the Cas9 protein is directed by gRNA to target specific genomic locus. This system allows multiplex genome editing, where multiple target gene sequences can be edited simultaneously in a single transfection reaction with the addition of multiple gRNAs. The system is versatile and simple to use, and changing target specificity only requires a change in the design of the GeneArt® CRISPR Strings™ DNA.

E-Learning: Bioreactors Made Easy!

The video instructions allow students and newcomers to fermentation to run laboratory experiments on their own. These instructions were compiled in co-operation with the University of Applied Sciences, Esslingen, (Germany) and are now available from INFORS HT.

The Cultivation Video Guide takes the user through the bioprocess step-by-step through short video clips and numerous photographs and diagrams. The guide accompanies the user through the entire bioprocess from the preparation of the pre-culture and steps to get the bioreactor ready to the actual fermentation process.

The idea was conceived by Prof. Dr. Richard Biener, University of Applied Sciences, Esslingen (Germany), since bioreactors can be damaged easily if handled inappropriately. "The operation of a bioreactor requires comprehensive training," according to Dr. Biener. "The students are excited about the instructions since complex operating steps can be shown much more comprehensibly in a video sequence". The instructions make the students more confident in the operation of the bioreactor allowing them to work more on their own and independently in the laboratory.

Dr. Burkhard Feigel (Managing Director Infors GmbH), confirms: "Younger users especially get detailed instructions without any preliminary training which allows them to perform their tasks more easily and without external help. The advisers see the special benefit in the fact that students and pupils make fewer operating errors and there are fewer defects. Basically, the instructions are very well-tried and proven in educational settings and are a type of support that is welcomed by advanced users as well."
