3D Culture Cell Harvesting Kit

The AMSBIO Cultrex® 3D Culture Cell Harvesting Kit is based on a non-enzymatic approach that prevents biochemical degradation during processing and overcomes the problems associated with extracting cells from 3D culture in BME or Laminin I.

The quality of the harvested cells is ideal for Western blotting and RNA and DNA analysis. In 3D culture, cells are grown in a microenvironment of specialized proteins that mimics in vivo conditions and allows the cells to organise into complex structures rather than uniform monolayers.



This new kit provides an optimised and standardised solution for the isolation and normalization of cell lysates from 3D Culture Matrix, BME or Laminin I for subsequent biochemical analysis.



3D Cultures exhibit cellular behaviours and morphologies similar to those seen in vivo; however, the adaptation of these models for studying biochemical processes has been impeded by the challenge of separating intact or 3D grown cells from extracellular proteins comprising the 3D matrix. 

Commonly, proteases are employed to degrade these extracellular proteins; however, proteases also degrade proteins on the cell surface, and protease activity may carry over into lysate preparations. Non-enzymatic methodologies have also been described for depolymerising extracellular matrix proteins, although the implementation of these protocols remains problematic for some researchers.

Affordably priced - Porvair flip cap centrifuge tubes can be spun up to 9400G and will withstand repeated autoclaving at 121C or long term storage at -80C. For easy identification each centrifuge tube has a solvent resistant writing area.

Porvair 15ml and 50ml flip cap centrifuge tubes have thin clear polypropylene walls and can withstand 8000G and 12000G respectively.

TAP Biosystems, a leading supplier of innovative cell culture systems and consumables for life science applications, is pleased to announce it is installing a system to automate the ex-vivo expansion of autologous cell therapies (CellBase CT™) at Czech-based firm PrimeCell, one of Central and Eastern Europe’s largest regenerative medicine companies.

The CellBase CT system at PrimeCell will be used in their new GMP facility at the National Tissue Centre Inc to rapidly scale up their manual cell culture processes in T-flasks and will provide a fully-contained aseptic cell expansion area. This will allow scientists to manufacture multiple autologous cell therapies from many individual patients in parallel and will enable the company to supply clients, including regional transplant centres in the Czech Republic, with a range of clean autologous therapies.

Joining the Optima line of floor and tabletop ultracentrifuges, this entry-level instrument offers context-sensitive onscreen help and is available in multiple native languages to meet a full range of separation requirements in a user-friendly environment.

Users can set multiple programmes of up to five steps each using an easily navigated, full-colour LCD touch-screen accessible in nine languages: English, Spanish, German, Japanese, French, Italian, Russian, Portuguese and Chinese (simplified).

Fully compatible with Beckman Coulter TL labware and fixed-angle, swinging bucket and unique near-vertical tube (NVT) rotors, the Optima MAX-TL can be used in applications including separation of subcellular particles, viruses and viral particles, rate-zonal separation of proteins in sucrose gradient, separation of lipoproteins, pelleting of RNA through a CsCl gradient, isopycnic separation of plasmid DNA and others.

With a maximum speed of 120,000 rpm and maximum rcf of 657,000 x g, the instruments control speed within ±50 rpm of set point.  Temperatures can be set from 0° to 40°C in 1°C increments, and controlled within ±2°C of set point (after equilibrium).  The units operate in ambient temperatures from 15° to 35°C.  Thermoelectric refrigeration eliminates CFCs and ODCs.

Beckman Coulter Optima MAX-TL ultracentrifuges fit in a standard BioSafety* hood, include HEPA filtration (filter available separately), and can use BioSafe* centrifuge tubes and fluid containment annulus rotors.  Operating sound level is <47dBa.

At this year’s Achema, Eppendorf and the DASGIP Information and Process Technology GmbH presented for the first time the new DASbox single-use vessel for parallel bioprocessing.

The development of the DASbox single-use vessel reinforces the strong synergy that benefits both DASGIP and the Eppendorf AG, which acquired the bioprocessing specialist this past January. “Within a very short time we were able to combine the long-standing competency of the Eppendorf AG in the field of plastics technology with our bioreactor expertise, which led to a new product”, explains Dr. Matthias Arnold, member of DASGIP management and head of the development department. 

The DASbox single-use vessel is the first single-use bioreactor which was developed for the DASGIP Parallel Bioreactor Systems. As a fully instrumented mini bioreactor with a working volume of 60-250 ml, it was constructed specifically for use in combination with the DASGIP DASbox mini bioreactor system. Especially with cell culture users in mind, the advantages of single-use technology are combined with the benefits of parallel cultivation and the full functionality of industrial bioreactors. All critical process parameters such as pH, soluble oxygen and optical density can be monitored and controlled using industry standard probes. 

Integrated dip tubes allow for controlled addition of liquids, sampling, as well as fully massflow controlled gas supply. The magnet coupled stirrer is only one of the examples for the uncompromising sterile technology of this single-use bioreactor.  A special feature of the DASbox single-use vessel is the novel, liquid-free Peltier element temperature control and condensation. This innovation, which enables, amongst others, efficient liquid condensation from the exhaust air, represents a further milestone in the transfer of the requirements of classic bioreactor technology to single-use technology.

The DASbox single-use vessel accelerates bioprocess development. The small working volumes save precious cell material and media, and extensive cleaning and sterilization procedures are superfluous. Less time is required for installation of the bioreactor, and cross contaminations are virtually excluded. Parallel cultivation under precisely controlled conditions delivers reliable results, fast. Further, the comprehensive software functions of DASGIP DASware allow user friendly and detailed capture, saving, analysis and management of resulting data.
